Mediphage was formed as a spin-off company from the University of Waterloo to commercialize a novel gene delivery vector technology - ministring DNA (msDNA).

Ministring DNA meets a critical need in the gene & cell therapy space. While gene and cell therapies hold great potential for the treatment of a wide range of chronic disorders, the costs remain astronomical, ranging from $400K to $1M per treatment. These high costs are due to the use and production of expensive vectors to deliver therapeutic DNA to cells. Additionally, significant safety issues exist in the approach; approximately 2,000 current gene therapy trials use viral vectors, owing to their superior efficacy. However, the safety risks associated with viral vectors, including insertional mutagenesis and immunogenicity, are barriers to making genetic therapies broadly applicable to many disease indications.

Role Description:

Mediphage Bioceuticals is recruiting a process development scientist to manage and direct Mediphage’s plasmid production and purification operations and development. The candidate will play a key role on the R&D team, overseeing and directing the development of large scale plasmid DNA manufacturing platforms. This role requires a highly-skilled, talented, and motivated scientist who will focus on the development, characterization, and scale up of methods for producing and purifying plasmid DNA. We seek candidates with proven ability to work independently after initial training and effectively collaborate to design and implement optimized project plans.

Essential Functions and Duties:

  • Oversee & direct optimization of critical scientific and engineering parameters for DNA production including bacterial cell culture and fermentation, harvest, feed strategies, and lysis and clarification
  • Chromatographic purification of DNA using SE, IEX, and HIC
  • Evaluate and oversee raw materials and equipment ensuring scalability and compatibility with GMP operations
  • Employ molecular techniques including DNA sequencing, restriction digest analysis, gel electrophoresis, small and large scale plasmid extraction, and generating glycerol stocks
  • Develop SOPs, batch records, and provide technical support and training to others
  • Develop supporting documentation for regulatory submissions
  • Assume responsibility for inventory management related to plasmid production and purification

Skills and Experience:

  • BS (6+), MS (3+), or PhD (1+) years industry experience. Degree in biochemistry, chemistry, chemical engineering, or molecular biology
  • Previous experience with biological production in bacterial culture systems
  • Industrial experience in the manufacturing of DNA products in fermentation bioreactors
  • Purification experience with size exclusion , anion exchange, or hydrophobic interaction chromatography
  • Ability to effectively manage and prioritize multiple parallel laboratory tasks is essential
  • Meticulous record keeping habits
  • Excellent interpersonal, written, and oral communication skills and ability to work as part of a multidisciplinary team
